The Track Matte Effect in After Effect might look a little intimidating and confusing when you use it the first time. But the concept behind that is pretty easy once you understand it.

If you don't see the Track Matte controls just unhide it with the Transfer Controls pane icon in the lower left.

Now you can set a Track Matte for any layer you want to mask. Just keep in mind that the mask layer must be right above this layer.

Here you have 4 options ...Alpha Matte + inverted....and Luma Matte + inverted.

That basically means....that you need alpha when transparency is involved in the mask layer...and luma when it's a black&white image.

the fact that the mask layer must be right above the masked layer can get pretty tedious when you need to mask multiple assets with the same mask layer again and again.

That's where the set matte effect comes in handy. With this effect you can target a specific mask in your composition and don't have to duplicate it over and over again. A great way to keep your composition nice and organized.
