Nathalia is the main damage dealer here to clear the first 2 waves with her AoE attacks and also to clear some Fire Imps around the boss, while Santis inflicts Poison on the boss to take down his health quickly. Both Mary and Ice Wolf are there to provide additional buff for Nathalia to deal extra damage through her unique trait.

The main important point to take away is Nathalia must have Ascension 2 to unlock the additional damage from her trait for every positive effect she has. Otherwise, you will encounter problems dealing sufficient damage to the waves and boss. Santis will need to have Ascension 5 to enable her trait to 100% land the Poison, so the run will be consistent.
