1. introduction to Pyrazinamide
It is weakly tuberculocidal and more active in an acidic medium.
2. Mechanism of Action
Pyrazinamide is an anti-tuberculosis drug that works by inhibiting the growth of mycobacteria, primarily by preventing the formation of a key component in their cell wall, mycolic acid. This leads to the accumulation of pyrazinoic acid in the bacteria, leading to the death of the bacteria.
3. Antibacterial Activity
Pyrazinamide is highly active against M. tuberculosis, particularly in the dormant or non-replicating stage of the bacterium. It is often used in combination with other antituberculosis drugs to enhance efficacy.
4. Mechanism of Resistance
Resistance to pyrazinamide can develop due to mutations in the pncA gene, which encodes an enzyme responsible for converting pyrazinamide to pyrazinoic acid.
5. Pharmacokinetics
Pyrazinamide is well absorbed after oral administration and is primarily metabolized in the liver. It has a half-life of approximately 4 hours and is excreted in the urine.
6. Side Effects
The most common side effects of pyrazinamide include nausea, vomiting, loss of
appetite, and arthralgia. In some cases, it can also cause liver toxicity, which may be serious and
even life-threatening. Other rare but serious side effects include rash, fever, and hypersensitivity
