Activated partial thromboplastin time (APTT) blood test: blood coagulation profile lecture 5

1. introduction
The test in which we measure the activity of intrinsic and common pathway of blood coagulation. Used to monitor the treatment effects with heparin i.e. anticoagulant.
2. principal
The APTT is based on the principle that in citrated plasma, the addition of a platelet substitute, factor XII activator, and CaCl2 allows for formation of a stable clot. The time required for the formation of a stable clot is recorded in seconds and represents the actual APTT result.

4. Manual method
Pipette 0.2 mL kaolin/platelet substitute mixture in small clean glass tube.
Add 0.1 mL of blood plasma and incubate at 37C in water bath for 2-3 min.
Add 0.1 mL 0.025 mol/l CaCl2 and start stopwatch
Hold the tube in water bath and tilt the mixture and observe clot formation.
Once the clot observe stop the stopwatch and note the time which is APTT.
5. Automated method using analyzer machine
On the machine
Deliver 50 μL of actine reagent and blood plasma in clean cuvette
Mix it will and keep it in machine at 37C for 3 min
Add and mix 50 μL of CaCl2 reagent.
After few second the result will be seen on screen
